RESUMO

High-rate activated sludge (HRAS) relies on good bioflocculation and subsequent solid-liquid separation to maximize the capture of organics. However, full-scale applications often suffer from poor and unpredictable effluent suspended solids (ESS). While the biological aspects of bioflocculation are thoroughly investigated, the effects of fines (settling velocity < 0.6 m3/m2/h), shear and surface overflow rate (SOR) are unclear. This work tackled the impact of fines, shear, and SOR on the ESS in absence of settleable influent solids. This was assessed on a full-scale HRAS step-feed (SF) and pilot-scale HRAS contact-stabilization (CS) configuration using batch settling tests, controlled clarifier experiments, and continuous operation of reactors. Fines contributed up to 25% of the ESS in the full-scale SF configuration. ESS decreased up to 30 mg TSS/L when bioflocculation was enhanced with the CS configuration. The feast-famine regime applied in CS promoted the production of high-quality extracellular polymeric substances (EPS). However, this resulted in a narrow and unfavorable settling velocity distribution, with 50% ± 5% of the sludge mass settling between 0.6 and 1.5 m3/m2/h, thus increasing sensitivity towards SOR changes. A low shear environment (20 s-1) before the clarifier for at least one min was enough to ensure the best possible settling velocity distribution, regardless of prior shear conditions. Overall, this paper provides a more complete view on the drivers of ESS in HRAS systems, creating the foundation for the design of effective HRAS clarifiers. Tangible recommendations are given on how to manage fines and establish the optimal settling velocity of the sludge.

BACKGROUND: Asymptomatic transmission was found to be the Achilles' heel of the symptom-based screening strategy, necessitating the implementation of mass testing to efficiently contain the transmission of COVID-19 pandemic. However, the global shortage of molecular reagents and the low throughput of available realtime PCR facilities were major limiting factors. METHODS: A novel semi-nested and heptaplex (7-plex) RT-PCR assay with melting analysis for detection of SARS-CoV-2 RNA has been established for either individual testing or 96-sample pooled testing. The complex melting spectrum collected from the heptaplex RT-PCR amplicons was interpreted with the support of an artificial intelligence algorithm for the detection of SARS-CoV-2 RNA. The analytical and clinical performance of the semi-nested RT-PCR assay was evaluated using RNAs synthesized in-vitro and those isolated from nasopharyngeal samples. RESULTS: The LOD of the assay for individual testing was estimated to be 7.2 copies/reaction. Clinical performance evaluation indicated a sensitivity of 100% (95% CI: 97.83-100) and a specificity of 99.87% (95% CI: 99.55-99.98). More importantly, the assay supports a breakthrough sample pooling method, which makes possible parallel screening of up to 96 samples in one real-time PCR well without loss of sensitivity. As a result, up to 8,820 individual pre-amplified samples could be screened for SARS-CoV-2 within each 96-well plate of realtime PCR using the pooled testing procedure. CONCLUSION: The novel semi-nested RT-PCR assay provides a solution for highly multiplex (7-plex) detection of SARS-CoV-2 and enables 96-sample pooled detection for increase of testing capacity. .

Background: Victorian Institute of Sports Assessment (VISA-A) is a patient-reported outcome for assessing symptoms severity associated with Achilles tendinopathy (AT). It is a valid and reliable tool that has been used widely for measuring and monitoring treatment outcomes for AT. This clinical measurement study aims to develop a Chinese version of the VISA-A questionnaire. The study objective is to adapt the VISA-A questionnaire cross-culturally and assess its psychometric property for Chinese-speaking individuals. Methods: VISA-A was translated and adapted cross-culturally according to international guidelines for self-reported questionnaires. During the establishment of Chinese VISA-A, there are 5 stages involved in the creation process, including translation, synthesis, reverse translation, review, and pretesting, which are performed by professionals in various fields, including orthopaedic surgeons, physiotherapists, and professional translators. Results: A total of 60 participants were recruited to complete the Chinese VISA-A and 36-Item Short Form Health Survey (SF-36) questionnaires. The overall test-retest reliability was 0.98 (intraclass correlation coefficient = 0.97-0.99). The correlation between Chinese VISA-A and physical function subscale (r = 0.70) was strong and statistically significant. There were moderate correlations between Chinese VISA-A, limitations to role of physical function subscale (r = 0.30), and bodily pain subscale (r = 0.42), which were also statistically significant. There were statistically significant differences in Chinese VISA-A scores between healthy control and pathologic group (P < .001), at-risk group, and pathologic group (P < .001) respectively. Conclusion: Chinese VISA-A demonstrated good reliability and validity for measuring symptom severity in patients with AT. Chinese VISA-A can be recommended as a self-reported measure for monitoring symptoms severity and treatment progress of patients with Achilles tendinopathy.Level of Evidence: Level II, cohort study.

Objective. To evaluate the use of an objective structured clinical examinations (OSCE) to assess clinical competency acquired during an off-campus introductory pharmacy practice experience (IPPE). Methods. Third-year pharmacy students completed an IPPE in transitions of care and completed 24 experiential contact hours at one of 17 practice sites. Students were assessed using two OSCEs, the first occurring prior to beginning an off-site IPPE (pre-experience OSCE) and the second occurring after completion of the off-site IPPE (post-experience OSCE). Each OSCE consisted of 10 stations and covered five graded competency domains. The primary outcome was the degree of change in student performance from the pre-experience OSCE to the post-experience OSCE. Secondary outcomes included changes in each graded domain, OSCE pass rate, and failure conversion rate. Results. Of 111 students, 109 completed both the pre- and post-experience OSCE. Significant improvements were observed in overall score and cohort pass rate. Overall scores improved from 80 for the pre-experience OSCE to 87 for the post-experience OSCE. The OSCE pass rate also improved from 47% to 84%. Conclusion. Although preceptor evaluations have traditionally served as the primary summative assessment for IPPE and APPE, this study indicates that OSCEs may be a reliable alternative to assess clinical competency acquired from off-site practice experiences.

RESUMO

This paper presents a generalization of the discrete z transform algorithm. It is shown that the GOD-ZT algorithm is a generalization of several important conventional discrete transforms. Based on the GOD-ZT algorithm, a tunable general optical discrete z transform (GOD-ZT) processor is synthesized using the silica-based finite impulse response transversal filter. To demonstrate the effectiveness of the method, the design and simulation of a tunable optical discrete Fourier transform (ODFT) processor as a special case of the synthesized GOD-ZT processor is presented. It is also shown that the ODFT processor can function as a real-time optical spectrum analyzer. The tunable ODFT has an important potential application as a tunable optical demultiplexer at the receiver end of an optical orthogonal frequency-division multiplexing transmission system.

Using a simplified chirp z-transform (CZT) algorithm based on the discrete-time convolution method, this paper presents the synthesis of a simplified architecture of a reconfigurable optical chirp z-transform (OCZT) processor based on the silica-based planar lightwave circuit (PLC) technology. In the simplified architecture of the reconfigurable OCZT, the required number of optical components is small and there are no waveguide crossings which make fabrication easy. The design of a novel type of optical discrete Fourier transform (ODFT) processor as a special case of the synthesized OCZT is then presented to demonstrate its effectiveness. The designed ODFT can be potentially used as an optical demultiplexer at the receiver of an optical fiber orthogonal frequency division multiplexing (OFDM) transmission system.

RESUMO

We show analytically and numerically that a practically realizable first-order optical Hilbert transformer (OHT) can simultaneously function as a negative/positive first-order temporal optical differentiator (OD) in the stopband and a broadband ±90° phase shifter in the passbands. An integrated-optic OHT based on a four-tap finite impulse response filter is designed using the Remez iteration algorithm and is numerically verified.

Inverse-Gaussian apodized fiber Bragg gratings (IGAFBGs) are numerically studied using the transfer matrix method and fabricated by the commonly used phase-mask scanning technique in a single-step scanning process. The IGAFBG can serve as a dual-wavelength passband filter, whose wavelength spacing can be continuously tuned by introducing a tunable chirp through applying a strain gradient in principle. Also, an IGAFBG with identical dual passbands having 0.144nm wavelength spacing is experimentally achieved. We also show that an IGAFBG can act as a multipassband filter with varied free spectral ranges (FSRs), and the largest FSR variation of this IGAFBG is nearly seven times more than that in a comparable FBG pair filter. An IGAFBG with varied FSRs of approximately 16.125, approximately 12.25, approximately 8.5, and approximately 6.375GHz is fabricated. This multipassband varying-FSR IGAFBG filter can find applications in step-tunable microwave generations.

A theoretical study of a new application of a simple pi-phase-shifted waveguide Bragg grating (PSWBG) in reflection mode as a high-speed optical dark-soliton detector is presented. The PSWBG consists of two concatenated identical uniform waveguide Bragg gratings with a pi phase shift between them. The reflective PSWBG, with grating reflectivities equal to 0.9, a free spectral range of 1.91 THz, and a nonlinear phase response, can convert a 40 Gbit/s noisy dark-soliton signal into a high-quality 40 Gbit/s return-to-zero signal with a peak power level of approximately 17.5 dB greater than that by the existing Mach-Zehnder interferometer with free spectral range of 1.91 THz and a linear phase response.

A novel universal support for deoxyribo- and ribonucleic acid synthesis has been developed. The support, constructed from 1,4-dimethoxycatechol, represents an improvement over existing universal supports because of its ability to cleave and deprotect under mild conditions in standard reagents. Because no nonvolatile additives are required for cleavage and deprotection, the synthesized oligonucleotides do not require purification prior to use in biochemical assays. Using reverse phase HPLC and electrospray mass spectroscopy, it was determined that oligonucleotides synthesized on the universal support (UL1) 3'-dephosphorylate quickly (9 h in 28-30% ammonium hydroxide (NH4OH) at 55 degrees C, 2 h in 28-30% NH4OH at 80 degrees C, or <1 h in ammonium hydroxide/methylamine (1:1) (AMA) at 80 degrees C). Oligonucleotides used as primers for the polymerase chain reaction (PCR) assay were found to perform identically to control primers, demonstrating full biological compatibility. In addition, a method was developed for sintering the universal support directly into a filter plug which can be pressure fit into the synthesis column of a commercial synthesizer. The universal support plugs allow the synthesis of high-quality oligonucleotides at least 120 nucleotides in length, with purity comparable to non-universal commercial supports and approximately 50% lower reagent consumption. The universal support plugs are routinely used to synthesize deoxyribo-, ribo-, 3'-modified, 5'-modified, and thioated oligonucleotides. The flexibility of the universal support and the efficiency of 3'-dephosphorylation are expected to increase the use of universal supports in oligonucleotide synthesis.

We present a theoretical study of a new application of a simple pi-phase-shifted fiber Bragg grating (PSFBG) in transmission mode as a high-speed optical temporal integrator. The PSFBG consists of two concatenated identical uniform FBGs with a pi phase shift between them. When the reflectivities of the FBGs are extremely close to 100%, the transmissive PSFBG can perform the time integral of the complex envelope of an arbitrary input optical signal with high accuracy. As an example, the integrator is numerically shown to be able to convert an input Gaussian pulse into an optical step signal.

What is believed to be a new approach for the design and analysis of a reconfigurable optical square pulse generator using the concept of temporal optical integration and the digital signal processing method is presented. The reconfigurable square pulse generator is synthesized using compact active semiconductor-based waveguide technology, and it consists simply of the cascade of a tunable microring resonator (or a tunable all-pole filter) and a tunable asymmetrical Mach-Zehnder interferometer (or a tunable all-zero filter). The reconfigurable generator can convert an input picosecond pulse (i.e., soliton or Gaussian pulse) into an optical square pulse. The pulse width of the generated square pulse can be adjusted by controlling the time delay of a variable delay element in the tunable all-zero filter. The reconfigurable generator can convert an input picosecond pulse train into return-to-zero (RZ) and non-return-to-zero (NRZ) signals with square pulse shapes. The repetition rates of the generated RZ and NRZ signals can be varied by adjusting the bit period of the input picosecond pulse train, the input pulse width, and the time delay of the variable delay element. The effect of the deviation of the parameter values on the generator performance is also studied.

The design and analysis of an Nth-order optical integrator using the digital filter technique is presented. The optical integrator is synthesized using planar-waveguide technology. It is shown that a first-order optical integrator can be used as an optical dark-soliton detector by converting an optical dark-soliton pulse into an optical bell-shaped pulse for ease of detection. The optical integrators can generate an optical step function, staircase function, and paraboliclike functions from input optical Gaussian pulses. The optical integrators may be potentially used as basic building blocks of all-optical signal processing systems because the time integrals of signals may sometimes be required for further use or analysis. Furthermore, an optical integrator may be used for the shaping of optical pulses or in an optical feedback control system.

We present a new hybrid optimization method for the synthesis of fiber Bragg gratings (FBGs) with complex characteristics. The hybrid optimization method is a two-tier search that employs a global optimization algorithm [i.e., the tabu search (TS) algorithm] and a local optimization method (i.e., the quasi-Netwon method). First the TS global optimization algorithm is used to find a "promising" FBG structure that has a spectral response as close as possible to the targeted spectral response. Then the quasi-Newton local optimization method is applied to further optimize the FBG structure obtained from the TS algorithm to arrive at a targeted spectral response. A dynamic mechanism for weighting of different requirements of the spectral response is employed to enhance the optimization efficiency. To demonstrate the effectiveness of the method, the synthesis of three linear-phase optical filters based on FBGs with different grating lengths is described.

RESUMO

We report, for the first time to our knowledge, the presence of concentration-induced nonuniform power in tunable erbium-doped fiber lasers. A theoretical model is proposed with pair-induced quenching taken into account. We obtain good agreement between numerical and experimental results of a high-concentration erbium-doped fiber ring laser with a large tuning range of over 100 nm. These findings are useful for the design of lasers with doped fibers.

An effective method for achieving a room-temperature multiwavelength erbium-doped fiber ring laser is presented. Simultaneous multiwavelength lasing with 0.5-nm intervals is achieved both experimentally and theoretically by addition of sinusoidal phase modulation in the ring cavity to prevent single-wavelength oscillation.

A simple reflow technique is employed for the fabrication of elliptical refractive microlens arrays (MLAs) on a low-cost inorganic-organic SiO2/ZrO2 sol-gel glass. The measured results show that the fabricated elliptical microlenses in a 256 x 512 array have excellent surface and dimensional qualities in terms of smoothness and uniformity. It is also shown that the optical parameters of the MLAs, such as the focal length and aperture dimension, can be accurately controlled. The proposed technique requires only an ordinary binary photomask for pattern transfer. Furthermore, the sol-gel material is found to be feasible for high-volume production, because the fabrication of MLAs can be implemented by use of cheap sol-gel materials without an etching step.

RESUMO

We report a widely tunable single-frequency linear-cavity erbium-doped fiber laser covering both the conventional wavelength band (C-band) and the long wavelength band (L-band). The laser has low threshold, high slope efficiency and high signal-to-noise radio. A large tuning range of over 100 nm is realized by optimization of the active fiber length.
